Tissue Prep Protocols

Block

5% NDS in 1xPBS containing ​

  • 0.6% Triton

  • 25% dimethyl sulfoxide

  • 0.01% azide

(overnight, RT)

Primary

​5% NDS in 1xPBS containing 

  • 0.8% Triton

  • 5% DMSO

  • 1:2000 Rabbit anti-NF

  • 1:100 Mouse anti-MBP

(5 days, RT)

Secondary

​5% NDS in 1xPBS containing 

  • 0.8 % Triton

  • 5% DMSO

  • 1:150 donkey anti-rabbit (488 or 568)

  • 1:150 donkey anti-mouse (488 or 568)

  • 1:1000 DAPI 

(7 days, RT)

Index Matching

  • 67% thiodiethanol in water

AcX

1:100 Acx in 1xPBS overnight

Gelation

47:1:1:1

Expansion

Autoclave or Proeteinase K

Storage

  • 1xPBS 

Prepare For Imaging

Polylysine

  • Coat bottom of 6-well plate (glass) with thin layer of polylysine

  • transfer gel to plate

Agar

​Fill plate with agar (PBS, TDE, or water)

Glass slide

Remove solidified agar from plate to large glass #2 coverslip

More Agar

Add more agar around puck to hold in place.

cover with small petri dish + saran wrap to avoid evaporation
